Increased Mrna Expression of Connexins and Transient Receptor Potential Channels in the Urothelium of Patients with Interstitial Cystitis: Possible Biomarkers

چکیده
Hypothesis / aims of study Interstitial cystitis (IC) is a disease characterized by bothersome lower urinary tract symptoms including bladder hypersensitivity, urinary frequency and bladder pain, which severely impair patient’s quality of life. However, pathophysiology of IC remains unclear despite the struggle for the development of new biomarkers to diagnose IC and effective therapeutic interventions. In the present study, to explore pathophysiology of IC, we investigated mRNA expression of connexions (Cx) and transient receptor potential (TRP) channels in the urothelium.

Localization and expression of inducible nitric oxide synthase in biopsies from patients with interstitial cystitis.
PURPOSE Interstitial cystitis is a chronic inflammatory disease of the bladder and luminal nitric oxide has been shown to be increased in the bladder in patients with interstitial cystitis. We analyzed endogenous nitric oxide formation and inducible nitric oxide synthase gene expression in the bladder of patients with interstitial cystitis to obtain further knowledge of the localization of indu...
متن کاملRole of CXCR2 and TRPV1 in functional, inflammatory and behavioural changes in the rat model of cyclophosphamide-induced haemorrhagic cystitis.
BACKGROUND AND PURPOSE Cyclophosphamide induces urotoxicity characterized by the development of cystitis, which involves bladder overactivity and inflammation. Here, we investigated the roles of chemokine receptor 2 (CXCR2) and transient receptor potential vanilloid 1 (TRPV1) channels in a rat model of cyclophosphamide-induced cystitis. EXPERIMENTAL APPROACH Cystitis induced by cyclophosphami...

OBJECTIVE To investigate the changes including expression and localization of 2 potassium channels, renal outer medullary K+ channel (ROMK) and voltage-gated K+ channel 7.1 (KCNQ1), after increased urinary potassium leakage in patients with interstitial cystitis/painful bladder syndrome (IC/PBS). MATERIALS AND METHODS The study group included 24 patients with IC/PBS and a control group consis...
متن کاملMorphine-induced analgesic tolerance is associated with alteration of protein kinase Cγ and transient receptor potential vanilloid type 1 genes expression in rat lumbosacral cord and midbrain
Introduction: Transient receptor potential vanilloid type 1 (TRPV1) and protein kinase Cγ (PKCγ) are involved in sensitization/desensitization to noxious stimuli. We aimed to examine the gene expression levels of TRPV1 and PKCγ in rat lumbosacral cord and midbrain on days 1, 4 and 8 of induction of morphine analgesic tolerance. Methods: Two groups of male Wistar rats received ...
متن کاملIntracerebroventricular Injection of Lipopolysaccharide Increases Gene Expression of Connexin32 Gap Junction in Rat Hippocampus
Introduction: Gap junctions are intercellular membrane channels that provide direct cytoplasmic continuity between adjacent cells. This communication can be affected by changes in expression of gap junctional subunits called Connexins (Cx). Changes in the expression and function of connexins are associated with number of brain neurodegenerative diseases. Neuroinflammation is a hallmark of vario...
